Datumetine Preferentially Upregulates NMDAR Signalling Pathways in Different Brain Regions of Mice

چکیده مقاله:
Background: We earlier reported that datumetine possesses binding affinity with NMDAR and that 14-day exposure to datumetine altered NMDAR signalling by mimicking glutamate toxicity. Here, we investigated the potential neuroprotective effect of a single shot of a low dose of datumetine administration in BALB/c mice. Results: Relative to Veh, datumetine downregulate the expression of CamKIIα in the hippocampus and PFC but not in the cerebellum, CREB was also upregulated in the PFC only, but pCREB was also upregulated in all the three brain regions observed, while BDNF was only upregulated in both hippocampus and PFC of Datumetine relative to Veh. MK-801 on the other hand reversed some of the effects of datumetine in the brain regions observed. No major histological alterations were observed in the different brain regions immunohistochemically. Conclusion: We conclude that a single low dose of datumetine moderately enhances NMDAR activity. This showed the neuroprotective potentials of low datumetine exposure.

Apoptosis: from Signalling Pathways to Therapeutic Tools
Apoptosis or programmed cell death is a gene regulated phenomenon which is important in both physiological and pathological conditions. It is characterized by distinct morphological features including chromatin condensation, cell and nuclear shrinkage, membrane blebbing and oligonucleosomal DNA fragmentation. Although, two major apoptotic pathways including 1) the death receptor (extrinsic) and...

1. The present study describes effects of extracellular ATP (ATPe) on plasma membrane potential and cytoplasmic Ca2+ concentrations ([Ca2+]i) in rat Sertoli cells. Sertoli cells in suspension were stimulated with ATPe and other nucleotides and ionic changes were monitored utilizing the fluorescent dyes bis-oxonol and fura-2/AM. ATPe induced a prompt plasma membrane depolarization which was depe...
متن کاملSignalling pathways mediating inflammatory responses in brain ischaemia.
Stroke causes neuronal necrosis and generates inflammation. Pro-inflammatory molecules intervene in this process by triggering glial cell activation and leucocyte infiltration to the injured tissue. Cytokines are major mediators of the inflammatory response. Pro-inflammatory and anti-inflammatory cytokines are released in the ischaemic brain. Anti-inflammatory cytokines, such as interleukin-10,...
متن کاملMapping glucose-mediated gut-to-brain signalling pathways in humans☆
OBJECTIVES Previous fMRI studies have demonstrated that glucose decreases the hypothalamic BOLD response in humans. However, the mechanisms underlying the CNS response to glucose have not been defined. We recently demonstrated that the slowing of gastric emptying by glucose is dependent on activation of the gut peptide cholecystokinin (CCK1) receptor. Using physiological functional magnetic res...
متن کاملGABA and Dopamine Release from Different Brain Regions in Mice with Chronic Exposure to Organophosphate Methamidophos
Organophosphates such as methamidophos, usually used in the agricultural field, have harmful effects on humans. Exposures to insecticides has been associated with many disorders, including damage to the central and peripheral nervous system. Chronic exposure to organophosphates may lead to persistent neurological and neurobehavioral effects. This study was conducted to determine the effect of m...
